I have a list of people I receive e-mail from and I would like to know what
is better: have a split entry for each person or create a complex regular
expression that matches all the e-mail addresses? The number of people
range from between 5 and 40 for a single split.

I don't think it matters much. Do whatever feels most comfortable to
you.

I am using a single not-so-complex regexp for the mails that my Berkeley
friends send me.

Both cases are unlikely to be significant compared to the time used
for disk I/O when splitting mail.
