With '!' you can mark a message. Is there a way that you can a list with
directories that have marked messages and also how many messages there
are in the directory?

This is not a direct answer, but it might help:

When I tikmark a message with '!', the folder containing that message
will show up in the Gnus Group buffer marked with an '*'.

That I know, but the 'problem' is that I have a lot of folders (some in
a closed topic) and I want to do programmatically something with the
info. For example I want to know which has more as a certain amount off
ticks, or a tick that is older as a certain amount off days, or ...
