I am trying to make a pointing finger the bullet point symbol.
When I put the bullet point into the document, it works just fine. But when defined as the pointer, it somehow isn't showing up.
What am I doing wrong?

You can separate the definition from the setup. Try

\defineitemgroup[prompt]

\setupitemgroup[symbol=bighand,style=italic]

The symbol is set before the value of the symbol-key is available to the 
environment and therefore never used. You can now either set the symbol 
with \setupitemize (or \setupitemgroup) before you call \startitemize 
(or \startitemgroup) or pass the symbol as keyword as shown below.

      \startitem
