Looks like lmtx adds extra vertical space after heads.
Compare those two MWE:
\setuppapersize [S4]
\setuphead [subject] [after={}]
\starttext
\startsubject[title={blabla}]
\input knuth
\stopsubject
\stoptext
\setuppapersize [S4]
\setuphead [subject] [after={}]
\starttext
\startcolumns [n=3]
\startsubject[title={blabla}]
\input knuth

\stopcolumns
\stoptext
