Move the \setlayer… command after \starttext or load a font with \setupbodyfont before you set the content.

\setvariables
  [address]
  [name=John Doe,
   street=21 Jumpstreet,
   zip=12345,
   town=TeX-City]

\definelayer[MyLayer][state=start]
\setupbackgrounds[page][background={MyLayer}]

\starttext

\setlayerframed[MyLayer][frame=off,align=right]
  {\getvariable{address}{name}\\
   \getvariable{address}{street}\\
   \getvariable{address}{zip} – \getvariable{address}{town}}

a

\stoptext
