Hi,
the colon sign disappears if one uses pxmath fonts (latest beta MkIV).
\setupbodyfont[palatino] % not OK
% \setupbodyfont[asana] % OK
\startTEXpage
$a:b$\par
$a\colon b$
\stopTEXpage
