Load a font or move the \setlayer commands after \starttext.

\setupbackgrounds[page][background=bkg]

\definelayer[bkg][x=0mm,y=0mm,width=\paperwidth,height=\paperheight]

\setupbodyfont[modern]

\setlayerframed[bkg][frame=off,width=\paperwidth,height=50mm]{\definedfont[Mono at 10mm]Some text}

\setlayerframed[bkg][frame=off,width=\paperwidth,height=\paperheight]{\definedfont[Mono at 240mm]X}

\starttext

\page[empty]

\stoptext
