What do you want to achieve with the line above because it doesn’t make any sense.
When you want to change the main font to Gentium you can do this with “\setupbodyfont[gentium]”.
\setupbodyfont[gentium]
\starttext
\startitemize
\startitem One \stopitem
\startitem Two \stopitem
\stopitemize
\stoptext
